(Euthanasia of animals used in research and teaching)

說明 實驗動物人道終止時機：動物實驗可能造成動物疼痛或緊迫，為維護動物福祉，研究人員應採取科學上合理最早可結束實驗的時機點，以求達到研究目的的同時，避免動物持續遭受不必要的疼痛與緊迫。除非IACUC審核同意以動物瀕死或死亡作為實驗終點，否則應在動物呈現痛苦、垂死、死後組織自體溶解、或死後被籠內其他同類啃食前，以人道的方式處理。 實驗預期的動物異常：在科學研究中，若動物的發病或死亡是可預期或必然的實驗後果，則研究者應考量研究需求、預期病程、最早可見之症狀指標等，檢討制定合適的人道終止時機，避免動物持續承受痛苦。 實驗導致的非預期異常：因實驗操作導致非預期的發病、瀕死、死亡、疼痛或持續的緊迫，動物須立即進行治療或安樂死。如治療會影響實驗結果，則動物需自實驗中移除並進行治療，或予以安樂死。如判斷此狀態為滿足研究目的所需，則必須修改動物實驗申請表，獲IACUC核准才可繼續執行。 自然發生的非預期異常：繁殖、實驗中或尚未使用之動物均可能發生與實驗無關之非預期異常。如發生非預期的瀕死、惡病質、無法攝食飲水等狀態，需立即安樂死。嚴重性較低但仍造成動物疼痛或緊迫的非預期狀況，則須評估對動物福祉及實驗結果的影響。如異常狀態會影響實驗結果，則動物需自實驗中移除或安樂死，如不影響實驗結果，則應給予治療。如治療會影響實驗結果，除非另經IACUC允許，否則動物需安樂死。有些動物模式如基因改造動物，可預期動物會自然發生損及動物福祉的異常，則應於動物實驗申請表中載明，以利動物照護人員發覺與判斷，給予合適的照護或使動物得到即時的處置。

泛用的人道終止時機如下。除IACUC已審查同意之情形(如：屬於實驗預期的異常，且能使動物痛苦程度減至最低)，否則只要符合下列任一項情況，就須將動物安樂死。

1.Body Condition Score (BCS)過低：評估後僅1分的動物須立即安樂死。評估為2分的動物如已能提供足夠實驗數據，須予以安樂死。小鼠的BCS詳如附圖，其他動物亦有對應之BCS可參考。

2.體重減輕(Weight loss)： 快速失去原體重的20-25%、或成長期動物持續無增重、未監測體重但動物呈現惡病質及持續性肌肉消耗時。非生長期動物體重減輕可依據動物剛進動物房之體重或平均年齡體重為依據。生長期之動物體重或許不會下降，但若無法正常增重，仍應判為體重減輕。 (Weight loss of 20-25% (depending on attitude, weight recorded at time of arrival, and age: growing animals may not lose weight, but may not gain normally) or if not measured, characterized by cachexia and wasting syndrome.)

3.食慾喪失(Inappetance)： 小型囓齒類動物完全喪失食慾達24-36小時或食慾不佳（低於正常量之50%）達3天時。大型動物完全喪失食慾達5 天或食慾不佳（低於正常量之50%）達7天時。 (Complete anorexia for 24 hours in small rodents, up to 5 days in large animals; partial anorexia (less than 50% of caloric requirement) for 3 days in small rodents, 7 days in large animals.)

4.虛弱（無法進食或飲水）(Weakness/inability to obtain feed or water)： 動物在沒有麻醉或鎮靜的狀態下，長達24小時無法站立或極度勉強才可站立時。 (Inability or extreme reluctance to stand, that persists for 24 hours, assuming that the animal has recovered from anesthesia.)

5.垂死/瀕死 (Moribund state)： 動物在沒有麻醉或鎮靜的狀態下，表現精神抑鬱伴隨體溫過低（低於37℃）時。(Depression coupled with body temperature below 37℃)

6.感染(Infection)： 呈現體溫上升或異常的血檢數值(如白血球數目增加)，對藥物治療(如抗生素)無良好反應且持續演變為全身性不適症狀出現時。 (Infection involving any organ system (either overt, or indicated by increased body temperature or WBC parameters), which fails to respond to antibiotic therapy within an appropriate period of time and is accompanied by systemic signs of illness.)

表2. 腫瘤試驗之動物安樂死時機與準則(Human Endpoint in Cancer Research):

說明 腫瘤模式動物的人道終止時機：在適當時機結束實驗可減少非特異性的系統變化影響，增進結果的準確度。設計實驗終點時應考量研究的特殊需求，例如腫瘤形成試驗，在見到明確的腫瘤生長時即可終止，但在致癌物質誘發皮膚乳突瘤的研究，細胞癌化可能發生在病程後期，因此實驗終點需設定在較晚的時間點。有許多方法如影像分析等，可以協助設立更精準的實驗終點，使終點盡可能提早，以符合人道精神。不允許以動物死亡或瀕死作為實驗終點。

接種腫瘤的實驗鼠必須至少每週觀察三次以上，以確保動物的生理狀況。觀察的項目包括：動物的外觀、姿態、行為、生理反應、食物飲水消耗、體重變化以及腫瘤的大小與外觀。 (Tumor-inoculated rodents must be observed at least three times a week for their physical conditions, which includes appearance, posture, behavior, physical reaction, food/water consumption, weight change, tumor size and tumor appearance.)

腫瘤潰瘍程度、腫瘤周圍組織變形程度，以及動物惡病質的程度，在設立人道的實驗終點時都需要納入考量。潰瘍是淺層組織壞死的傷口，可能呈現乾燥、化膿或有滲出液，假如潰瘍壞死深度穿透皮膚，或是持續出現滲出液超過48小時，則動物應接受安樂死。

腫瘤負荷：在取得所需研究數據的前提下，實驗設計應將每隻動物的腫瘤負荷控制在最低限度。舉例如下： 1.功效測試研究：當統計數據顯示出可信且顯著之療效，就該終止實驗。 2.治療藥物的研究：應從設計上避免發生需要讓控制組腫瘤生長過大的狀況。 3.單純用於例行繼代的腫瘤，應制定大小上限。 4.在一般腫瘤研究中，動物生長單顆腫瘤時，不該讓腫瘤平均直徑生長超過20mm(小鼠)及40mm(大鼠)。 5.如動物身上複數位置接種腫瘤，則所有腫瘤體積相加不應超過前述單顆時的腫瘤體積。 6.在自發產生腫瘤之動物或腫瘤誘發實驗，可能生長多顆腫瘤，除非有堅強的科學理由，其腫瘤負荷的加總不應超過前述單顆的上限。 7.原位腫瘤、淋巴網狀內皮系統的腫瘤，及轉移性腫瘤等，較難評估腫瘤負荷，可藉由前驅實驗找出其動力學特性、分布模式，以利預測臨床症狀之發展，並設立合適的人道終點。

腫瘤生長的位置會影響可允許的腫瘤負荷程度，腳掌、尾巴、眼球和骨頭等位置較易引起疼痛，因此需要足夠的科學理由支持，並制定較早的實驗終點。在腦腫瘤研究，可配合MRI或螢光等影像分析，結合體重變化情形，將體重減輕作為實驗終點的指標。

除上列表一安樂死準則外，若接種腫瘤動物出現以下臨床症狀時，即為humane endpoints，應立即安樂死該實驗動物。 (Animal inoculated with tumor cells should be euthanized if following conditions is observed.) A.腫瘤生長超過動物原體重的10%。平均腫瘤直徑在小鼠超過20mm、在大鼠超過40mm。多顆腫瘤之體積相加超過前述上限。 (Tumor weight exceed 10% of the animal weight. Average diameter of tumors over 20mm in mice or 40mm in rats.)
